About: Palonosetron

Palonosetron belongs to a drug class called 5HT3 receptor antagonists, used for the prevention and treatment of nausea and vomiting, particularly that caused by chemotherapy, radiation therapy, or postoperatively. The use of Palonosetron, compared with traditional 5-HT3 Ra’s, can be more effective in preventing chemotherapy-induced nausea and vomiting (CINV) during the first 24 hours following chemotherapy, which is the leading cause of premature discontinuation of chemotherapy. Palonosetron has the longest elimination half-life in its class of drug, after administration.

INDICATIONS

Chemotherapy

Palonosetron is used to prevent acute or delayed nausea and vomiting that occurs within and after the first 24 hrs of chemotherapy. Palonosetron is used in ongoing courses of chemotherapy regimens that have a 30 to 40% chance of causing nausea and vomiting, known as, moderately emetogenic cancer chemotherapy (MEC). Children as young as 1 month old can take Palonosetron.

Postoperative

Palonosetron is used as a post-operative nausea and vomiting (PONV) preventative. PONV is a common unpleasant adverse reaction of surgery and general anesthesia. Palonosetron has the highest binding harmony to the 5-HT3 receptor out of its drug class, with an elimination half-life of approximately 40 hours that provides extended relief to patients receiving the drug.

Upset Stomach

Palonosetron is also used to prevent upset stomach and throwing up, caused by food poisoning, alcohol poisoning, and symptoms of infection in the stomach or other organs. Palonosetron is also prescribed for reasons not listed.
